RESUMO

Through microorganism in the rumen of ruminant, plant fiber can be converted to edible food such as meat and milk. Ruminants had a rich and complex microbial community within the rumen, and the bacteria comprised the dominant proportion of the ruminal microbes. High-throughput sequencing offered a viable solution for the study of rumen microbes. In this study, rumen fluid samples were taken from 11 cattle from Inner Mongolian, the DNA of 11 rumen fluid samples were extracted and bacterial amplicons of the V4 regions of 16S rRNA were subjected to Illumina sequencing. More than 90,000 raw reads and 60,000 effect Tags per sample were obtained. 28,122 operational taxonomic units (OTUs) were observed from 11 samples, in average 2557 ± 361 OTUs for each sample. Bacteroidetes (44.41 ± 7.31%), Firmicutes (29.07 ± 3.78%), and Proteobacteria (7.18 ± 5.63%) were the dominant phyla among the bacteria of rumen, accounting for 82%. At the genus level, the highest relative abundance was Prevotella. Their functions were predicted using the Kyoto Encyclopedia of Genes and Genomes (KEGG). The results showed that they included metabolism, genetic information processing, environmental information processing and cellular processes. It explored the bacterial community diversity and composition of the rumen of Mongolian cattle. On the whole, our research showed that there was a high diversity as well as rich bacterial flora function of rumen bacteria in Mongolian cattle. Meanwhile, these findings provided information for further studies on the relationship between the community, diversity, functions of rumen bacteria and the nutritional physiological functions of the host.

This study monitored 20 organophosphate esters (OPEs) in water and sediment from three typical mariculture bases (Yunxi Marine Ranching (YX), Hangzhou Bay (HZB), and Zhelin Bay (ZLB)) and Meiliang Bay (MLB) of Taihu Lake in China, focusing on the spatial distribution and sources of OPEs. Moreover, the occurrence and risk of OPEs in fishes from ZLB were evaluated. The ∑OPE concentrations in waters followed the order MLB (591 ng/L) > YX (102 ng/L) > HZB (70.0 ng/L) > ZLB (37.4 ng/L), with tri(1-chloro-2-propyl) phosphate (TCIPP), triethyl phosphate (TEP), and tri(2-chloroethyl) phosphate (TCEP) being the dominant OPEs. Significantly higher ∑OPE concentrations were found in sediment in MLB compared to the other three areas with similar levels. The decreasing concentrations of OPEs from nearshore to offshore areas in HZB and MLB indicated that terrigenous input is the main source of OPEs. The even distribution of OPEs in YX and ZLB combined with PCA analysis suggested ship traffic or aquaculture activities are also potential sources. The ∑OPE concentrations in fishes ranged from 0.551-2.45 ng/g wet weight, with TCIPP, tri-phenyl phosphate (TPHP), and TCEP being the main OPEs. Hydrophobicity was a key factor affecting the sediment-water distribution coefficients and the bioaccumulation factors of OPEs. The human exposure to OPEs through consumption of fishes from ZLB had a low health risk.

ETHNOPHARMACOLOGICAL RELEVANCE: In traditional Chinese herbal medicine, rhubarb is said to remove accumulation with purgation, clearing heat, and discharging fire. Modern pharmacology has shown that rhubarb extract has a purgative effect when given to experimental animals in an appropriate dose. However, the active components and their mechanism of action are still not clearly defined. AIM OF THE STUDY: The current research aimed to evaluate the synergistic stool-softening effects and explore the action mechanism of rhubarb free anthraquinones (RhA) and their monomers on constipation in rats. MATERIALS AND METHODS: A rat model of water deficit-induced constipation was established to induce constipation, and these rats were treated with RhA and its monomers. ELISA, histopathology, immunohistochemistry, qPCR and Western blotting based on network pharmacology and molecular docking were conducted to explore the possible mechanism of action of RhA and its monomers. RESULTS: RhA, aloe-emodin, rhein, and chrysophanol showed stool-softening activity, and the combination of aloe-emodin and rhein had the strongest softening effect on faecal pellets. Aloe-emodin, rhein, and chrysophanol significantly increased the serum levels of vasoactive intestinal peptide (VIP), motilin (MTL), and substance P (SP), upregulated the expression of VIP, cyclase-associated protein 1 (CAP1), protein kinase A (PKA), cystic fibrosis transmembrane conductance regulator (CFTR), aquaporin 3 (AQP3), aquaporin 4 (AQP4), and aquaporin 8 (AQP8), decreased the expression of epithelial sodium channel (ENaC) and Na+/H+ exchanger 3 (NHE3), and reduced the colonic tissue concentration of Na+-K+-ATPase in the constipated rats. Osmolality of colonic fluid in model rats treated by RhA, aloe-emodin, rhein, and chrysophanol was increased. CONCLUSION: Aloe-emodin, rhein, and chrysophanol were the stool-softening components of the RhA extract, and there were certain drug-interactions between the components. RhA upregulated VIP expression, activated the cyclic adenosine monophosphate protein kinase A (cAMP/PKA) pathway, and further stimulated CFTR expression while inhibiting NHE3 and ENaC expression, resulting in a hypertonic state in the colonic lumen. Water transport could then be driven by an osmotic gradient, which in turn led to the upregulation of AQP3, AQP4, and AQP8 expression. In addition, RhA likely improved gastrointestinal motility by increasing serum VIP, SP, and MTL concentrations, thus promoting faecal excretion.

Overproduction of lactate (LA) can occur during exercise and in many diseases such as cancers. Individuals with hyperlactatemia often display anemia, decreased serum iron, and elevated hepcidin, a key regulator of iron metabolism. However, it is unknown whether and how LA regulates hepcidin expression. Here, we show LA binds to soluble adenylyl cyclase (sAC) in normal hepatocytes and affects systemic iron homeostasis in mice by increasing hepcidin expression. Comprehensive in vitro, in vivo, and in silico experiments show that the LA-sAC interaction raises cyclic adenosine monophosphate (cAMP) levels, which activates the PKA-Smad1/5/8 signaling pathway to increase hepcidin transcription. We verified this regulatory axis in wild-type mice and in mice with disordered iron homeostasis. LA also regulates hepcidin in humans at rest and subjected to extensive exercise that produce elevated LA. Our study links hyperlactatemia to iron deficiency, offering a mechanistic explanation for anemias seen in athletes and patients with lactic acidosis.

Various unique spatial structures are often found in the enzymes of biological systems. From the consideration of bionics, it is challenging but meaningful to design nanozymes with distinctive structures to enhance their bioactivities. To explore the relationship between the structure and activity of nanozymes, in this work, a special structural nanoreactor, namely small pore black TiO2 coated/doped large pore Fe3O4 (TiO2/-Fe3O4) loaded with lactate oxidase (LOD), was constructed for chemodynamic and photothermal synergistic therapy. Specifically, LOD loaded on the surface of the TiO2/-Fe3O4 nanozyme alleviates the low level of H2O2 in the tumour microenvironment (TME); the black TiO2 shell with multiple pinhole channels and a large specific surface area not only facilitates LOD loading, but also enhances the affinity of the nanozyme for H2O2; H2O2 is continuously enriched on the surface of the TiO2/-Fe3O4 nanozyme and transmitted to mesoporous Fe3O4, in turn efficiently producing abundant toxic hydroxyl radicals (˙OH) for chemodynamic therapy. Meanwhile, the TiO2/-Fe3O4 nanozyme under 1120 nm laser irradiation has excellent photothermal conversion efficiency (η = 41.9%), and further accelerates the production of ˙OH for amplifying the chemodynamic therapy efficiency. This self-cascading, special structure nanozyme provides a novel strategy for application in highly efficient tumour synergetic therapy.

BACKGROUND: Organophosphate esters (OPEs) are common endocrine-disrupting chemicals, and OPE exposure may be associated with type 2 diabetes (T2D). However, greater knowledge regarding the biomolecular intermediators underlying the impact of OPEs on T2D in humans are needed to understand biological etiology. OBJECTIVES: We explored the associations between OPE exposure and glycometabolic markers among older Chinese adults 60-69 years of age to elucidate the underlying mechanisms using a multi-omics approach. METHODS: This was a longitudinal panel study comprising 76 healthy participants 60-69 years of age who lived in Jinan city of northern China. The study was conducted once every month for 5 months, from September 2018 to January 2019. We measured a total of 17 OPEs in the blood, 11 OPE metabolites in urine, and 4 glycometabolic markers (fasting plasma glucose, glycated serum protein, fasting insulin, and homeostatic model assessment for insulin resistance). The blood transcriptome and serum/urine metabolome were also evaluated. The associations between individual OPEs and glycometabolic markers were explored. An adverse outcome pathway (AOP) was established to determine the biomolecules mediating the associations. RESULTS: Exposure to five OPEs and OPE metabolites (trimethylolpropane phosphate, triphenyl phosphate, tri-iso-butyl phosphate, dibutyl phosphate, and diphenyl phosphate) was associated with increased levels of glycometabolic markers. The mixture effect analysis further indicated the adverse effect of OPE mixtures. Multi-omics analyses revealed that the endogenous changes in the transcriptional and metabolic levels were associated with OPE exposure. The putative AOPs model suggested that triggers of molecular initiation events (e.g., insulin receptor and glucose transporter type 4) with subsequent key events, including disruptions in signal transduction pathways (e.g., phosphatidylinositol 3-kinase/protein kinase B and insulin secretion signaling) and biological functions (glucose uptake and insulin secretion), may constitute the diabetogenic effects of OPEs. DISCUSSION: OPEs are associated with the elevated risk of T2D among older Chinese adults 60-69 years of age. Implementing OPE exposure reduction strategies may help reduce the T2D burden among these individuals, if the relationship is causal. https://doi.org/10.1289/EHP11896.

The occurrence of per- and polyfluoroalkyl substances (PFAS) was investigated inside two manufacturing facilities in China. Levels, profiles, and spatial distribution of the detected PFAS were found to be distinctly site-specific and influenced by the area's historic function, production structure of the plant, downpour-induced accidental pollution, and variations in the adsorption and transport of compounds. Very high concentrations of PFAS [mainly C4 and C8 perfluoroalkyl sulfonic acids (PFSAs)] were found in topsoil and groundwater from both plants, with the highest values of 4.89 × 106 µg/kg dw and 1.10 × 104 µg/L, respectively. Elevated concentrations of perfluoroalkyl carboxylic acids (PFCAs) in this study were attributed to their unintentional formation during the electrochemical fluorination process, which might be an overlooked source of PFCA. PFAS generally showed decreasing trends from shallow layers to the bottom of the soil core and demonstrated some downward migrations at different soil depths with time, and C4-C8 PFAS presented a deeper seepage than their long-chain homologues. Total organic carbon appeared to be more important for PFAS sorption to the topsoil than to the soil core. Workers were at potential risk of exposure to perfluorooctanesulfonic acid via soil at production and storage related sites. This study provides a critical reference for the systematic control of PFAS pollution around manufacturing facilities and a proof for an overlooked source of PFCA.

Per- and polyfluoroalkyl substances (PFASs, n=22), including emerging alternatives, in dust samples were analyzed using high-performance liquid chromatography-tandem mass spectrometry (HPLC-MS/MS) to study their pollution characteristics. These samples were collected from main and minor roads in Shijiazhuang. Some of the roads were located near sewage treatment plants and fire stations. The results showed that PFASs were ubiquitous in the road dust of Shijiazhuang; in particular, hexafluoropropylene oxide dimer acid (HFPO-DA), an alternative, was measured for the first time in China. The total concentrations of PFASs ranged from 2.62 to 137.65 ng·g-1. Perfluorooctanoic acid (PFOA) was the dominant PFAS, followed by perfluorobutanoic acid (PFBA), HFPO-DA, and perfluorooctane sulfonic acid (PFOS). The highest and lowest levels of PFASs were observed in the northwest and southeast regions of Shijiazhuang, respectively. The compositions of PFASs were obviously different in road dust near sewage treatment plants and fire stations, especially for the types of emerging alternatives. Health risk assessment indicated that road dust intake had a low risk of human exposure to PFASs and emerging alternatives. Among the three routes (ingestion intake, inhalation intake, and dermal contact), ingestion intake was the main route for PFASs and emerging alternatives in road dust to enter the human body. Under the same exposure route, the exposure dose of children was higher than that of adults.

Coastal lagoons provide many important services to human society, but their year-round use for aquaculture introduces large amounts of sewage. The contamination of antibiotic resistance genes (ARGs) is therefore of great concern. In this study, 50 ARGs subtypes, two integrase genes (intl1, intl2), and 16S rRNA genes were detected by high-throughput quantitative PCR, and standard curves of all target genes were prepared for quantification. The occurrence and distribution of ARGs in a typical coastal lagoon (XinCun lagoon, China) were comprehensively explored. We detected 44 and 38 subtypes of ARGs in the water and sediment, respectively, and discuss the various factors influencing the fate of ARGs in the coastal lagoon. Macrolides-lincosamides-streptogramins B was the primary ARG type, and macB was the predominant subtype. Antibiotic efflux and antibiotic inactivation were the main ARG resistance mechanisms. The XinCun lagoon was divided into eight functional zones. The ARGs showed a distinct spatial distribution owing to the influence of microbial biomass and anthropogenic activity in different functional zones. Fishing rafts, abandoned fish ponds, the town sewage zone, and mangrove wetlands provided a large quantity of ARGs to the XinCun lagoon. Nutrients and heavy metals also significantly correlated with the fate of the ARGs, especially NO2--N and Cu, which cannot be ignored. It is noteworthy that lagoon-barrier systems coupled with persistent pollutant inputs result in coastal lagoons acting as a "buffer pool" for ARGs, which can then accumulate and threaten the offshore environment.

Thyroid disrupting chemicals (TDCs) have received much attention due to their potential adverse effects on animal and human health, which calls for rapid screen assays to identify them. The triiodothyronine (T3)-induced Xenopus metamorphosis assay (TiXMA) we developed previously has been successfully applied to the detection of the TDCs disrupting thyroid hormone (TH) signaling. Here, we attempted to expand the application of the TiXMA to the screening of the TDCs interfering with the hypothalamic-pituitary-thyroid (HPT) axis. Two well-known TH synthesis inhibitors methimazole (MMI) and sodium perchlorate (SP) were employed to test the sensitivity of the TiXMA to the TDCs interfering with the HPT axis. As expected, we observed that the two chemicals concentration-dependently antagonized T3-induced morphological changes and body weight reduction of X. laevis tadpoles following 96 h-exposure, in parallel with blocked thyroid development and down-regulated tshß expression in the brain. All the data show that both MMI and SP exert inhibitory effects on T3-induced metamorphosis, indicating that the TiXMA is capable of screening the TDCs interfering with the HPT axis. In comparison with Amphibian Metamorphosis Assay (AMA), a 21-day assay for screening the TDCs interfering with the HPT axis, the TiXMA has a remarkable advantage of shorter exposure duration (96 h).

A vital bioactive component of marine resources is Holothuria leucospilota polysaccharides (HLP). This study examined whether HLP could regulate intestinal flora to treat loperamide-induced constipation. Constipated mice showed signs of prolonged defecation (up by 60.79 min) and a reduced number of bowel movements and pellet water content (decreased by 12.375 and 11.77%, respectively). The results showed that HLP treatment reduced these symptoms, reversed the changes in related protein expression levels in the colon, and regulated the levels of active peptides associated with the gastrointestinal tract in constipated mice, which significantly improved water-electrolyte metabolism and enhanced gastrointestinal motility. Meanwhile, it was found that intestinal barrier damage was reduced and the inflammatory response was inhibited through histopathology and immunohistochemistry. As a means to further relieve constipation symptoms, treatment with low, medium, and high HLP concentrations increased the total short-chain fatty acid (SCFA) content in the intestine of constipated mice by 62.60 µg/g, 138.91 µg/g, and 126.51 µg/g, respectively. Moreover, an analysis of the intestinal flora's gene for 16S rRNA suggested that the intestinal microbiota was improved through HLP treatment, which is relevant to the motivation for the production of SCFAs. In summary, it was demonstrated that HLP reduced loperamide-induced constipation in mice.

Compound fruit drink (CFD) is a functional drink prepared with fruit, Chinese herbs and prebiotic fructooligosaccharide as the main ingredients. Loperamide hydrochloride was used to establish a mouse model of constipation. And the effect of CFD on the improvement of constipation and the impact on gut microbiota were studied. The results showed that CFD significantly enhanced intestinal motility in constipated mice (P < 0.05). It significantly improved serum levels of gastrointestinal regulatory-related peptides, elevated the short-chain fatty acids (SCFAs) content and alleviated colonic injury. Meanwhile, CFD also up-regulated the mRNA expression levels of AQP3, AQP9, SCF and c-Kit and the related protein expression levels. Fecal microbial results showed that the CFD medium-dose group significantly increased species richness. Furthermore, CFD increased the abundance of potentially beneficial bacteria and reduced the number of potentially pathogenic bacteria. This study indicated that CFD was a promising functional drink for effectively relieving constipation.

Per-and polyfluoroalkyl substances (PFASs) are omnipresent globally and received increasing attention recently. However, there are limited data on PFASs in the Tibetan Plateau (TP), a remote high-altitude mountain region, which is regard as an important indicator region to study long-range transport behaviors of contaminants. This study investigates the occurrence, distribution, partitioning behavior, and sources of 26 PFASs in water and sediments from the four lakes of TP. The ΣPFAS concentrations ranged from 338 to 9766 pg L-1 in water, and 12.2-414 pg g-1 dry weight in sediments. Perfluorobutanonic acid (PFBA) and perfluorooctane sulfonate (PFOS) were detected in all samples. Qinghai Lake had the highest ΣPFAS concentrations in both water and sediments, while the Ranwu Lake had the lowest. The functional groups and CF2 moiety units were investigated as essential factors influencing the partition behavior. Principal component analysis (PCA) combined back-trajectory was used to infer possible sources of PFASs. The results suggested that the main source of PFASs in Yamdrok Lake, Namco Lake, and Ranwu Lake on southern TP were mainly originated from South Asia via long-range atmospheric transport (LRAT); while for the Qinghai Lake of northern TP, LRAT, local emissions, and tourism activities were the primary sources of PFASs.

It is extremely important to analyze the contaminative behaviors of Perfluoroalkyl acids (PFAAs) due to their serious threats to urban environments which are closely related to humans. Current study aimed to explore the distribution, source apportionment and ecological risk assessment of PFAAs in surface water from Shijiazhuang, China. The concentrations of ∑PFAAs ranged from 19.5 to 125.9 ng/L in the investigation area. Perfluorobutanesulfonic acid (PFBS) and perfluoropentanoic acid (PFPeA) were the predominant contaminants (mean value: 14.3 ng/L and 16.6 ng/L, respectively). The distribution of PFAAs according to geospatial analysis and hierarchical clustering analysis (HCA) showed that higher levels of ∑PFAAs were detected in the southern surface water of Shijiazhuang and there was a stepwise decrease from the wet season to the dry season. Furthermore, based on source apportionment, the dominant potential sources were found to be wastewater treatment plant (WWTP) effluents and industrial discharge. The risk quotients (RQs) revealed low ecological risks of all PFAAs for aquatic organisms in Shijiazhuang surface water. Collectively, this study provided basic data for regulatory strategies for controlling PFAA pollutions in urban surface water.

Organophosphorus pesticides (OPPs) have raised an increasing public concern due to their harmful impacts. To explore the occurrence and distribution of OPPs in southeastern China marginal seas (SCMS), a sampling campaign was carried out from East China Sea (ECS) to South China Sea (SCS). A total of 33 OPPs are quantified with the ΣOPPs concentrations ranging from 4.73 to 14.15 ng/L. Higher ΣOPPs concentrations in the surface seawater from the estuaries of Yangtze River, Minjiang River, and Pearl River than those at other sampling sites indicates that riverine emissions are the principal sources of OPPs in SCMS. Different compositions of OPPs in ECS and SCS highlight the different priority of use categories for OPPs in China coastal region. In addition, the vertical diffusion and upwelling ocean currents play critical roles in the redistribution of OPPs in SCMS. For the first time, the ΣOPPs mass inventories in surface seawater of ECS and SCS are estimated at 8.51 and 11.26 t, respectively. Although the current ecological risk of OPPs is at low level in surface seawater of SCMS, the long-term use and bio-accumulative potential point to the necessity for the normalized monitoring of OPPs in China.

As the available building blocks are limited, improving the structural complexity of covalent organic frameworks (COFs) is still challenging. In this work, a new three-component COF synthesis strategy is established by using 4-aminobenzaldehyde as the bifunctional linker. The lattice parameters, crystallinity, and porosity of these COFs can be efficiently tuned by varying the amount of this linker. This work provides a new way to expand the structural diversity and complexity of imine-linked COFs.

Pan-fried green tea (PGT) is an easily acceptable tea drink for general consumers. In this study, volatile profiles and characteristic aroma of 22 representative Chinese PGT samples were extracted using stir bar sorptive extraction (SBSE) and analysed by gas chromatography-mass spectrometry (GC-MS), gas chromatography-olfactometry (GC-O) analysis, and odour activity value (OAV) calculations. In total, 88 volatile compounds were identified. Alcohols (45%), esters (19%), and ketones (16%) were the dominant volatiles, and geraniol (484.8 µg/kg) was the most abundant volatile component in PGT, followed by trans-ß-ionone and linalool. In addition, the differences of aroma characteristics among PGT and other three types of green tea, namely baked green tea, steamed green tea, and sun-dried green tea, were also observed using partial least squares discriminant analysis (PLS-DA) and heatmap analysis, and it was found that ß-myrcene, methyl salicylate, (E)-nerolidol, geraniol, methyl jasmonate were generally present at higher content in PGT. This is the first comprehensive report describing the volatile profiles of Chinese PGT, and the findings from this study can advance our understanding of PGT aroma quality, and provide important theoretical basis for processing and quality control of green tea products.

To study a high-efficiency online classroom intelligent management system, this article builds an artificial intelligence classroom management system based on the tensor CS reconstruction model. Moreover, this study uses the cosine function to represent the data energy fitting of the traditional active contour model and proposes a local cosine fitting energy active contour model based on partial image restoration, which is used for image and composite image segmentation. Simultaneously, this study proposes a new type of super-resolution algorithm. This algorithm performs Fourier transform of a low-resolution image into a frequency range and then performs an inverse Fourier transform on the image expanded in the frequency range to obtain the initial high-resolution image and finally reconstructs a new super-resolution image using the frequency-domain compressed data of the high-resolution image. Finally, this study verifies and analyzes the performance of the model through experiments. The research results are basically consistent with the expectations of the model.

Anisotropic nanoparticles possess high sensitivity to the environmental refractive index that is strongly dependent on their active facets. As a polyhedron exclusively enclosed by active facets, Au nano-rhombic dodecahedra (AuNRDs) with multiple tips and edges have been less explored with respect to their capability in surface plasmon resonance (SPR) sensing. Here, we report on the use of AuNRDs in the colorimetric detection of S2- and Hg2+. Upon Ag coating, the AuNRDs can probe S2- with high sensitivity and selectivity due to the chemical transformation of Ag into Ag2S. Furthermore, Hg2+ can be detected by the SPR of the AuNRDs based on the formation of HgS from the Ag2S shell on the AuNRDs. These changes in the morphology and composition of the AuNRDs have been confirmed by electron microscopy and elemental mapping. The promising colorimetric response of Ag-coated AuNRDs to S2- is visible through the naked eye and the change in the SPR of the AuNRDs shifted linearly with an increasing concentration of S2- in the range of 0.5-15 µM, with a detection limit of 0.26 µM. The change in the SPR of the Ag2S-coated AuNRDs also indicates a dependence on the concentration of Hg2+ in the range from 0 to 100 µM, with a detection limit of 38 nM. The limits of detection are lower or close to the maximum allowable levels of S2- and Hg2+ for drinking water defined by national or international administrations. These seminal results unveil new opportunities for the AuNRDs and other anisotropic nanostructures with active facets in SPR-based sensing applications.
